We report the identification of a recurrent, 520-kb 16p12.1 microdeletion associated with childhood developmental delay. The microdeletion was detected in 20 of 11,873 cases compared with 2 of 8,540 controls and replicated in a second series of 22 of 9,254 cases compared with 6 of 6,299 controls. Most deletions were inherited, with carrier parents likely to manifest neuropsychiatric phenotypes compared to non-carrier parents. Probands were more likely to carry an additional large copy-number variant when compared to matched controls. The clinical (...) features of individuals with two mutations were distinct from and/or more severe than those of individuals carrying only the co-occurring mutation. Our data support a two-hit model in which the 16p12.1 microdeletion both predisposes to neuropsychiatric phenotypes as a single event and exacerbates neurodevelopmental phenotypes in association with other large deletions or duplications. Analysis of other microdeletions with variable expressivity indicates that this two-hit model might be more generally applicable to neuropsychiatric disease. © 2010 Nature America, Inc. Welchen Standards müssen Personen genügen, damit ihre Äußerungen als wahrhaftig angesehen werden? Nicht Wahrhaftigkeit als soziale Tugend und auch nicht Wahrhaftigkeit als notwendige Argumentationsvoraussetzung , sondern die normativen Voraussetzungen wahrhaftiger Äußerungen von Überzeugungen, Werten, Wünschen und Gefühlen einer Person, d. h. eine Analyse der normativen Erfordernisse der Wahrhaftigkeit als Teilaspekt einer Sprachpragmatik, sind das Thema dieses Aufsatzes. Ausgehend von Überlegungen Wittgensteins, John Broomes und Robert Brandoms untersuche ich die normative Feinstruktur der Differenz zwischen dem 'Äußern' und dem 'Besitzen' eines propositionalen (...) Gehalts sowie der Vorsätzlichkeit dieser Differenz im 'Äußern'. BackgroundApproximately 7% of the general population is affected by an orphan disease, which, in the United States, is defined as affecting fewer than 1 in 1500 people. Disease awareness is often low and time-to-diagnosis delayed. Different legislations worldwide have created incentives for pharmaceutical companies to develop drugs for orphan diseases. A journalistic article in Bloomberg Businessweek has claimed that pharmaceutical companies have tried marketing orphan drugs by placing a specific disease into the popular television series “House, M.D.” which features diagnostic (...) journeys and was produced between 2004 and 2012. This study aimed to describe the presentation of orphan diseases in the television series “House, M.D.”, to test in an exploratory fashion the hypothesis that treatable orphan conditions are overrepresented in “House, M.D.” and to discuss whether such marketing practices may or may not be ethical.MethodsA list of all medical cases depicted in the television series “House, M.D.” was obtained and classified as orphan or non-orphan according to the Orphanet database. The ratios of orphan diseases among all diseases, such with an orphan drug designation and such with an orphan drug approval by the FDA were then compared with conservative approximations of real world conditions (chi-squared tests for equality of proportions). STROBE criteria were respected.ResultsOut of a total of n = 181 different medical diagnoses, n = 42 (23.2%) were orphan diseases. The difference in percentages in between “House, M.D.” and reality was not statistically significant for orphan diseases overall (p = 0.96), yet was statistically significantly higher for both orphan diseases with one or more orphan drug designations (p = 0.0192) and such with one or more approved orphan drugs (p < 0.0001).ConclusionsOrphan diseases with a designated and/or approved orphan drug were overrepresented in the television series “House, M.D.” with statistical significance while orphan diseases overall were not. This may be explained by (so far) undocumented efforts of pharmaceutical companies to place their orphan drugs in the television series, as described in the article in Bloomberg Businessweek. In his retelling, all (...) claims to glory for Homer's heroes are undermined; a world that is presented by Homer as brutal but honorable is replaced, by Shakespeare, with one that is rife with social and physical disease. As I continued to consider why Shakespeare displayed such hostility toward the Iliad, I found myself recognizing multiple, much more subtle, allusions to the Trojan War in another of his plays set in ancient Greece, The Comedy of Errors (COE). In time, it became apparent that the connection of these two plays with each other, and with the Iliad, was deliberate; through this comparison, Shakespeare rejects the Homeric ethos in favor of a Christian one. A consideration of René Girard's work on Shakespeare helps to bring home the connection between the plays and the concern that they illuminate.In his Theatre of Envy, Girard famously claimed that Shakespeare was consciously familiar with the concepts of mimetic desire and its relationship [End Page 183] to collective violence.1 He drew upon T&C and COE for examples: T&C for its explication of the importance of degree, its employment of collective murder, and the association of mimetic contagion with disease,2 and COE primarily for its emphasis on the special role of twins in mimetic theory as exemplars of identical rivals.3 Girard, I believe, is correct that Shakespeare understood the mechanisms and implications of mimetic theory, and he demonstrates how in these plays its consequences play out in a purely pagan world, and then in another in which there is a saving Christian intervention.The intellectual current of Shakespeare's time has been characterized by two primary themes: Reformation and Renaissance. The use of the word reformation, when taken on its own terms, suggests a benign and necessary process. But for an Englishman in the late 16th century, reformation also would have meant violence, displacement, fracture, war, and the disintegration of a once-unified Christendom. This crumbling of the ancient universal catholic church was accompanied by the Renaissance, a term used to refer to the rebirth of interest in, and progressive obsession with, the culture and literature of the ancient Greeks and Romans.4 My argument is that the two plays discussed here can be read as Shakespeare's response to these developments, demonstrating an anxiety over the threat of resurgent pagan values to a troubled Christianity, with T&C's ending standing as a warning of the danger of a return to the scapegoat mechanism, and COE reminding the reader of the power of the Christian alternative.The essay begins with a brief reminder of the depth of the enthusiasm for the classical era that emerged in Europe in the century leading up to, and including, the life of Shakespeare. The paper introduces a conceptually important, but previously unknown essay by the Russian poet, theologian and philosopher Aleksey Khomyakov. This essay, “The Slavic and Orthodox Christian Population of Austria,” was discovered in two versions: an original, previously unpublished manuscript and a later anonymous 1845 text. The author reveals an aesthetic function that certain structural elements perform in Khomyakov’s essay, encouraging the interaction between historiosophical ideas and literary creativity. The essay is emphatically philosophical in its style, as its very composition embraces (...) the classic Hegelian triad of “Being—Nothing—Becoming.” This notion overlaps with Khomyakov’s historiosophical idea of the European Slavdom that flourished in the first millennium B.C., and that then lost its sovereignty being under German rule, and finally enjoyed a renaissance in the 19th century. The paper also shows that the distinctive feature of Khomyakov’s poetry is its prophetism. The contrastive analysis was carried out to identify the affinity between the thematic-figurative patterns of Khomyakov’s early poetry and his essay on the Austrian Slavs, which revealed how the poetic thought had been accumulating and what aspect it had taken on within a philosophical and historical discourse. The author of the article puts forward the hypothesis that the difference in depicting Peter the Great in the two versions of Khomyakov’s essay can be attributed to his dispute with Konstantin Aksakov’s poem “To Peter.” This polemic between two fellow-Slavophils was driven by their diverging approaches to the problem of the ethical dilemma of ends and means. This unpublished manuscript of the Spanish Dominican Domingo Báñez reflects his personal account of the proceedings held during July 1602 in Valladolid in defense of his own doctrine against suspicious theses formulated by some Jesuits from Alcalá de Henares the previous March. The Jesuits denied that the adhesion of faith to the Roman Pontiff included him as a specific man, e.g. Pope Clement VIII. In support of their thesis, they provided the authority of Báñez. The Dominican theologian clarified in Valladolid (...) that the fact that Clement VIII was Pope did belong to the faith indeed but not in a primary and immediate sense, although this was later supported by other Jesuits in a third session held a few days later, again in Valladolid. Dieser Leon erblickte um 939 das Licht der Welt. Es ist möglich und sogar wahrscheinlich, läßt sich aber nicht mit absoluter Sicherheit sagen, ob er der ältere und damit erstgeborene der beiden Brüder war. Der Altersunterschied der beiden Brüder war jedenfalls nicht allzu groß. Aus einem Brief ihres Vaters geht hervor, daß beide Brüder in ihrer frühen (...) Kindheit anscheinend relativ schwer erkrankt waren. Es ist möglich, aber doch eher unwahrscheinlich, daß gerade diese, in dem Brief erwähnte Erkrankung zum Tode Leons führte. Leon war möglicherweise noch gegen Ende der Herrschaft des Kaisers Romanos I. Lakapenos, im Jahre 944 am Leben. Er starb aber vermutlich in diesem Jahr oder zu Beginn des nächsten Jahres. Am 6. April 945, zum Zeitpunkt der Mitkaiserkrönung seines Bruders Romanos, weilte er jedenfalls nicht mehr unter den Lebenden. Es ist möglich, aber nicht zu beweisen, daß er in dem im Zeremonienbuch erwähnten kleinen Larnakion im Heroon Konstantins des Großen in der Apostelkirche zu Konstantinopel, der Begräbnisstätte seiner Familie, beigesetzt war. Bohmian mechanics is a realistic interpretation of quantum theory. It shares the same ontology of classical mechanics: particles following continuous trajectories in space through time. For this ontological continuity, it seems to be a good candidate for recovering the classical limit of quantum theory. Indeed, in a Bohmian framework, the issue of the classical limit reduces to showing how classical trajectories can emerge from Bohmian ones, under specific classicality assumptions. In this paper, we shall focus on a technical problem that (...) arises from the dynamics of a Bohmian system in bounded regions; and we suggest that a possible solution is supplied by the action of environmental decoherence. However, we shall show that, in order to implement decoherence in a Bohmian framework, a stronger condition is required rather than the usual one. (shrink)

The original proof of the four-color theorem by Appel and Haken sparked a controversy when Tymoczko used it to argue that the justification provided by unsurveyable proofs carried out by computers cannot be a priori. It also created a lingering impression to the effect that such proofs depend heavily for their soundness on large amounts of computation-intensive custom-built software. Contra Tymoczko, we argue that the justification provided by certain computerized mathematical proofs is not fundamentally different from that provided by surveyable (...) proofs, and can be sensibly regarded as a priori. We also show that the aforementioned impression is mistaken because it fails to distinguish between proof search (the context of discovery) and proof checking (the context of justification). By using mechanized proof assistants capable of producing certificates that can be independently checked, it is possible to carry out complex proofs without the need to trust arbitrary custom-written code. We only need to trust one fixed, small, and simple piece of software: the proof checker. This is not only possible in principle, but is in fact becoming a viable methodology for performing complicated mathematical reasoning. This is evinced by a new proof of the four-color theorem that appeared in 2005, and which was developed and checked in its entirety by a mechanical proof system. (shrink)

Ma (...) poiché la prematura scomparsa dello studioso, al quale Maisano riconosce «appassionata adesione alla materia» e «signorile onestà intellettuale» (I, p. 75), impedì che l'opera venisse portata a termine, la presente risulta essere la prima edizione criticamente rivista, tradotta e commentata in modo completo: oltre ai 59 contaci di sicura attribuzione, essa comprende in appendice anche alcuni carmina dubia (83, 84, 87 Maas-Trypanis), nonché l'Inno acatisto, che sicuramente non può essere assegnato al Melodo (II, p. 580), ma del quale è ristampata la traduzione di Carlo Del Grande (Firenze 1948), proprio per documentare «la differenza anche stilistica fra questo componimento e i cantici di Romano» (II, p. 581). This article is a commentary on ChatGPT and LLMs (Large Language Models) in general. It argues that this technology has matured to the point where calling systems such as ChatGPT “stochastic parrots” is no longer warranted. But it also argues that these systems continue to have serious limitations when it comes to reasoning. These limitations are much more severe than commonly thought. A large array of examples are given to support these claims.

In this paper, we tackle the widely held view that practice-oriented approaches to ethics are conservative, preserving the moral status quo, and, in particular, that they do not promote any change in our dealings with animals or formulate clear principles that help us to achieve such change. We shall challenge this view with reference to Wittgensteinian ethics. As a first step, we show that moral thought and action rest on basic moral certainties like: equals are to be treated equally and (...) unequals unequally. We then explore the relations between this insight and the notion of the difficulty of reality elaborated by Cora Diamond. Our goal is to show that a Wittgensteinian-Diamondian approach to animal ethics is not necessarily morally conservative. Indeed, it offers a profound practice-oriented approach to animal ethics that is capable of promoting change in human dealings with animals because it is compatible with radical critique. (shrink)

This paper unifies different branches in the literature and points to promising research directions, whilst also providing an accessible introduction to key concepts in statistics and machine learning – as far as they are concerned with reliability.

In the famous exchange between Passennus Paulus and Javolenus Priscus at Plin. Ep. 6.15, it has not been previously recognized that Priscus’ reply is metrical and carries on the hexameter begun by Paulus. This opens up some interesting new possibilities for the interpretation of the letter.

The article attempts to compare personalist aspects in the works of Vasily Zenkovsky and Vladimir Lossky. It is shown that two types of philosophical personalism (metaphysical and existentialist) in the history of Russian thought set the framework for two types of theological personalism presented respectively by Zenkovsky and Lossky. The philosophy of Lev Lopatin was the important source for the principles of Zenkovsky’s personalist vision. The relevant philosophical background on Lossky’s personalism is provided by Nikolai Berdyaev’s works. The article considers (...) the deep criticism by the French theologian Jean-Claude Larchet, who seeks to demonstrate philosophical and existentialist sources of Lossky’s theological personalism, rather than patristical ones. It is noted that the criticism addressed to Lossky’s theory may slide to challenging the very possibility of an Orthodox theological personalism as a line of thought. It is shown that Zenkovsky’s works, presenting a nonexistentialist type of personalism, do not provoke such criticism. It’s concluded that whereas Lossky’s legacy in its personalistic aspect is challenged, Zenkovsky’s personalism enables another way of conceiving “Orthodox theological personalism.”. (shrink)

PA is Peano Arithmetic. Pr(x) is the usual Σ1-formula representing provability in PA. A strong provability predicate is a formula which has the same properties as Pr(·) but is not Σ1. An example: Q is ω-provable if PA + ¬ Q is ω-inconsistent (Boolos [4]). In [5] Dzhaparidze introduced a joint provability logic for iterated ω-provability and obtained its arithmetical completeness. In this paper we prove some further modal properties of Dzhaparidze's logic, e.g., the fixed point property and the Craig (...) interpolation lemma. Diamonds Moralphilosophie stellt dem einen praxissensitiven Ansatz gegenüber, der Emotionen und die moralische Imagination umfasst, wobei Diamond die Bedeutung des Menschseins betont.
